چکیده

In order to elucidate further the possible role of specific cytokines in the pathogenesis of atopic dermatitis (AD) the in vitro production of interleukin 4 (IL-4) and interferon-gamma (IFN-gamma) in patients with severe atopic dermatitis (n = 4) was compared with that in a group of non-atopic healthy controls. Overall IL-4 production by PHA- and PWM-driven PBMNCs was increased in controls during the first 48 h in culture. Addition of interleukin 2 (IL-2) into parallel cultures generated an insignificant (p > 0.05) increase in IL-4 production in AD patients compared with that from controls. IFN-gamma production by PWM-stimulated PBMNCs was markedly decreased in AD patients compared with controls (p < 0.01). Addition of IL-2 (250 U/ml) to parallel cultures failed to restore IFN-gamma production in AD patients. Finally, no IL-4 or IFN-gamma activity could be detected in any of the sera. In conclusion, the data suggest a possible dysregulation of cytokine production in at least a subgroup of AD patients, with an impaired capacity to secrete IFN-gamma, but a partially intact IL-4 generating capacity.

Different lymphocyte markers and cytokine expression in peripheral blood mononuclear cells in children with acute atopic dermatitis.

BACKGROUND The pathogenesis of atopic dermatitis (AD) is still not completely understood. AD is characterized by the presence of clinical symptoms of both IgE antibody-mediated immediate hypersensitivity and specific T lymphocyte-mediated delayed hypersensitivity. OBJECTIVE To evaluate the immunological mechanisms involved in children with acute AD lesions. MATERIAL AND METHODS Ten children...

Wrestlers’ immune cells produce higher interleukin-6 and lower interleukin-12 and interleukin-13 in response toin vitro mitogen activation

Objective(s):Although recent investigations have shown chronic inflammation and inflammation-associated diseases might be ameliorated by exercise; little is known about the relation between exercise training with anti/pro-inflammatory cytokines. Materials and Methods: This cross sectional study was conducted to compare interleukin-4 (IL-4), IL-6, IL-10, IL-12, IL-13, interferon gamma (IFN-γ) le...

In vitro Effect of Pomalidomide on Bone Marrow Mononuclear Cells from Multiple Myeloma Patients

Background: Many features of anticancer drugs, including cytotoxicity and/or cytokine induction, are studied using cell lines orhuman blood leukocytes. However, in a disease such as multiple myeloma, most cancerous cells are resided within bone marrowmononuclear cells. In the present study, we investigated the effect of pomalidomide on apoptosis and IL-2 production of bonemarrow mononuclear cel...

Evaluation of effective factors in the optimization of immunization to achieve antibodies against RhD antigen in culture medium

Abstract Background and Objectives Antibody against RhD antigen is important in blood transfusion and clinical medicine. Therefore, in this study, the effective factors in the optimization of in vitro immunization to achieve immunized lymphocytes that produce antibodies were evaluated to obtain antibodies against RhD antigen in the culture medium. Materials and Methods In an experimental study...

Suppression of IFN-gamma production in atopic group at the acute phase of RSV infection.

Several studies have suggested that respiratory syncytial virus (RSV) bronchiolitis induced the change of cytokine production profile in childhood. We sought to determine whether the RSV-induced cytokine production was affected by the patient's atopic background. We quantified interferon-gamma (IFN-gamma) and interleukin (IL)-4 in the supernatant of peripheral blood mononuclear cells (PBMCs) cu...
